چکیده
Alkaline proteases are the most important group of industrial enzymes that hydrolyse peptide bond proteins into small peptides. The demand alkaline protease predominantly from microbial origin has been recently increased and enhances research for with high stability at extreme conditions. Thus, this study is aimed to characterize bacterial isolate, Bacillus subtilis VBC7 screened dairy waste dumped soil. Extracellular production was carried out in broth by submerged fermentation. medium 10% ground nut extract pH 10 40 °C enhanced activity (712 U/mL) than other wastes such as coconut pulp sesame seed extract. This optimized media growth rate compared unsupplemented basal medium. Further, partially purified assessed their molecular weight (~30 k Da) 12 % SDS -PAGE. enzyme observed stable over a wide range (5-12), temperatures (10-70 °C). In addition, stimulated presence Mg2+, Ca2+ Mn2+ unpretentious after treating surfactants (SDS, Triton X-100, Tween 80), organic solvents (ethanol, methanol, chloroform, acetone hexane) inhibitors (EDTA β-mercaptoethanol). These compatible features could lead potential candidate among sectors.
